bFGF对脑出血大鼠海马组织SOD活力、MDA含量和Bax、Bcl-2转录水平的影响

摘    要:目的:观察碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)对大鼠脑出血模型出血侧海马组织中SOD活力、MDA含量和Bax、Bcl-2转录水平的影响。方法:采用脑内注射胶原酶建立大鼠脑出血模型,将模型鼠分为模型组、NS组和bFGF组各54只,模型组不作任何干预,bFGF组肌肉注射bFGF(8ug/kg,1次/天),NS组肌肉注射等量生理盐水。各组分别于干预第1、3、7天取出血侧海马组织,检测SOD活力、MDA含量及Bax mRNA、Bcl-2 mRNA的表达水平。结果:与NS组、模型组比较,bFGF组出血侧海马组织第7天SOD活力明显增高、MDA含量明显下降(P〈0.05);第3、7天BaxmRNA表达水平明显下降、Bcl-2 mRNA表达水平明显增高(P〈0.05)。结论:bFGF可通过促进氧自由基清除、降低Bax/Bcl-2基因的表达比值抑制细胞凋亡,从而保护脑出血后脑损伤。

Abstract:Objective:To study the effects of basic fibroblast growth factor(bFGF)on the activity of SOD,MDA content and expressions of Bax and Bcl-2 genes in rat hippocampus with intracerebral hemorrhage(ICH).Methods: The experimental ICH rat models were established by injecting collaginase Ⅶ into the internal capsule and randomly divided into ICH,normal saline(NS) and bFGF treated groups.The activity of SOD and content of MDA in hippocampus were measured,and the expression levels of Bax and Bcl-2 genes in hippocampus were evaluated by RT-PCR.Results: After bFGF intervention,SOD activity in the hippocampus in the bFGF group was significantly increased when compared with the NS control group P<0.05),whereas the MDA contents were significantly decreased(P<0.05).The expression of Bax gene in the bFGF group was lower than that in the NS control group(P<0.05),while the expression of Bcl-2 gene in the bFGF group was increased significantly(P<0.05).Conclusion: bFGF could provide a neuroprotective effect by eliminating free radicals,improving SOD activity and suppressing neural apoptosis.
