| 摘 要： | 目的构建A型流感病毒核蛋白（NP）基因部分序列的原核表达质粒。并诱导表达谷胱甘肽巯基转移酶融合蛋白。方法采用PCB方法扩增NP基因部分序列，并定向插入原核表达载体pGEX-4T-1中。转化大肠杆菌BL21．将阳性菌落经IPTG诱导目的基因融合蛋白的表达。结果扩增到NP基因部分序列，构建成重组表达质粒pGEX—S NP,并在大肠杆菌中进行了表达。结论成功构建A型流感病毒NP部分基因序列的原核表达质粒，并在大肠杆菌中表达了目的基因融合蛋白。

| Abstract: | Objective To construct an expressional plasmid encoding the partial sequence of Influenza A virus Nucleoprotein(NP) and express GST-S_(NP) fusion protein. Methods The partial sequence of NP gene was amplified by PCR and inserted into pGEX-4T-1 vector.Then the recombinant plasmid was transformed into E.coli BL21,identified by colony PCR and confirmed by DNA sequencing.The expression of target gene was induced with IPTG. Results The partial sequence of NP gene was amplified by PCR and inserted into pGEX-4T-1 properly.The recombinant fusion protein was expressed in BL21. Conclusion The recombinant plasmid pGEX-S_(NP) has been successfully constructed and recombinant protein of the target gene is expressed in E.coli BL21. |
