咖啡因对链尿霉素诱发大鼠β-细胞损伤的保护作用

 目的 探讨咖啡因对链尿霉素(streptozotocin,STZ)诱导大鼠β-细胞损伤的影响.方法 Wistar大鼠皮下注射生理盐水(对照组)或咖啡因(治疗组).15 min后,大鼠再皮下给柠檬酸缓冲或STZ(65 mg/kg).注射后第3天对大鼠进行口服葡萄糖耐量试验(OGTT).OGTT后第3天分离胰腺测定其中胰岛素水平.结果 STZ处理的大鼠,与对照组比较,在OGTT测试所有时间点都明显增加血浆葡萄糖水平.预先给咖啡因可明显减低ST2的升高血糖作用.STZ处理还可明显减少血浆胰岛素的浓度,但预先给咖啡因任何剂量不能逆转血浆胰岛素降低的作用.与对照组比较,STZ处理可明显降低胰腺中胰岛素水平.此作用也能被咖啡因所逆转.结论 此研究结果表明咖啡因对STZ损伤大鼠胰岛β-细胞,具有一定的保护效应.但要了解咖啡因对2型糖尿病患者β-细胞是否有保护效果尚需进一步研究.
