高压氧预处理对高原肺水肿大鼠肺水通道蛋白-5的影响

摘    要: 目的 观察高压氧预处理( hyperbaric oxygen preconditioning,HBO -PC)对高原肺水肿(high - altiude pulmonary edema,HAPE)大鼠肺水通道蛋白-5(NQP5)的影响.方法 38只Wistar大鼠,分对照组、HAPE模型组(HAPE组)及高压氧预处理组(HBO组)制作HAPE模型后,分别以Western Blot法及Real - Time PCR法观察各组大鼠肺AQP5蛋白、基因表达,同时观察肺水含量及病理变化.结果 HAPE组肺湿/干比明显高于Control组;HBO组肺损伤积分明显低于HAPE组;HAPE组AQP5及AQP5mRNA较Control组显著降低,HBO组AQP5及AQP5mRNA均明显高于HAPE组;肺组织损伤与肺AQP5表达有密切的相关关系,且呈负相关 结论 AQP5表达降低可能参与了HAPE的发病机制;HBO - PC能上调AQP5表达,具有预防和减轻HAPE的作用
