摘    要:目的研究豆腐柴中总黄酮的最佳提取工艺及其抗菌作用效果。方法以提取时间、料液比、乙醇浓度分数作为自变量,以豆腐柴中总黄酮(按芦丁计)的提取率作为因变量,采用响应面法优选总黄酮的最佳提取工艺,并采用紫外分光光度法测定豆腐柴中总黄酮的含量。结果确定最优提取工艺为提取时间为25.95 min,料液比为1∶24.05,乙醇浓度为81.89%,提取次数为3次,在此条件下,豆腐柴中总黄酮得率理论上可达0.807%。结论星点设计-效应响应面法优选豆腐柴中总黄酮类的提取工艺,方法简单,结果可靠。

Abstract:Objective To optimize extraction techniques for total flavonoids from Premna microphylla Turcz. by using central composite design and response surface method. Methods With the extraction time, solvent ratio, ethanol con-centration served as independent variables while the total extracted flavonoid (by rutinum) served as the dependent variable. With the central composite design and response surface method were employed to optimize the extracting process, and determinated the contents of total flavonoids premna by UV method. Results The optimized extraction conditions showed that, the best conditions were extraction time of 25.95 minutes, the ratio of solvent to material of 1: 24.05, ethanol concentration of 81.89% and extracted of 3 times, under these conditions, Premna microphylla Turcz. total flavonoids yield could up to 0.807% theoretically. Conclusion The central composite design and response surface method is simple in methodology and yet yields reliable results.
