氟比洛芬酯预处理对切口痛大鼠痛行为及中脑导水管周围灰质C-fos蛋白表达的影响

摘    要:目的通过研究不同剂量氟比洛芬酯预处理对切口痛大鼠痛行为及中脑导水管周围灰质C-fos蛋白表达的影响,探讨氟比洛芬酯的预防性镇痛效果。方法 30只健康成年雄性SD大鼠,随机分为5组,每组6只。模型组(S组)采用Brennan法进行手术,建立切口痛模型;对照组(C组)于术前20min尾静脉注射生理盐水1ml。F1(小剂量组)、F2(中剂量组)、F3(大剂量组)组分别于术前20min按1mg/kg、5mg/kg、9mg/kg尾静脉注射氟比洛芬酯。连续观察1h行为学反应,采用累积疼痛评分法进行评分,术后2h处死动物,取脑组织,分离中脑进行免疫组化染色,观察大鼠中脑导水管周围灰质内C-fos蛋白的表达情况。结果与S组和C组比较,F1、F2、F3组的累积疼痛评分较低且C-fos免疫阳性细胞的数量减少(P〈0.01);F1、F2、F3组随着给药剂量的增大,镇痛效果增强。结论氟比洛芬酯预处理可以减轻切口痛大鼠的术后疼痛,有预防性镇痛作用。

Abstract:Objective To observe the analgesic effect of Flurbiprofen axetil pretreatment on incisional pain in rats by injecting different doses of flurbiprofen axetil before making model,discuss the preventive analgesic effect of flurbiprofen axetil.Method 30 healthy adult male SD rats were randomly divided into 5 groups(n=6),model group(S group):the rat model was made according to the Brennan method,control group(C group):20min before the operation,rats were injected normal saline by 1ml;F1(low dose group),F2(middle dose group),F3(high dose group):20min before the operation,rats were injected flurbiprofen by 1mg/kg,5mg/kg,9mg/kg.Behavioral responses were observed for 1h,use cumulative pain score test to evaluate the pain behavior.2 hours later,kill the rats,isolate brain tissue,use immunohistochemistry method to observe the C-fos expression in rat midbrain periaqueductal gray.Results Compared with the S group and C group,the cumulative pain score of F1,F2,and F3 group was lower and the expression of C-fos decreased(P0.05);F1,F2,and F3 group,with the dose increased,the analgesic effect was enhancing.Conclusion Flurbiprofen axetil can reduce postoperative pain in incisional pain rats.
