肿瘤坏死因子相关的凋亡诱导配体联合顺铂对小鼠移植型肝癌抑制作用的研究

摘    要:目的探讨肿瘤坏死因子相关的凋亡诱导配体(TRAIL)联合顺铂(DDP)对小鼠移植型肝癌的抑制作用。方法 H22肝癌移植瘤小鼠随机分为生理盐水(对照)组、TRAIL组、TRAIL+DDP组和DDP组,称取瘤重分析其抑瘤作用、流式细胞检测细胞凋亡率。结果 (1)与对照组相比,TRAIL、DDP对小鼠移植性肝癌生长均有明显的抑制作用(P〈0.01),TRAIL与DDP联用有增效作用(P〈0.05)。TRAIL组、DDP组及TRAIL+DDP组的抑瘤率分别为45.14%、57.98%和77.23%。(2)与单一用药组相比,TRAIL与DDP联用可明显提高肿瘤细胞的凋亡率。结论 TRAIL与DDP联用对小鼠H22肝癌移植瘤的生长具有协同抑制作用。

Abstract:Objective To study the inhibitory action of TRAIL combined with cisplatin(DDP) on the transplanted hepatocellular carcinoma in mice.Methods Mice bearing H22 hepatocellular carcinoma were randomly divided into normal saline(NS) group,TRAIL group,TRAIL combined DDP group and DDP group.The tumor weight changes of mice were observed.The apoptotic rate was calculated by flow cytometry.Results(1) Compared with the control group,both TRAIL and DDP could inhibit the growth of the transplanted HCC in mice(P 〈 0.01) .They have a synergistic effect(P 〈 0.05) .The inhibitory rates in TRAIL,DDP and TRAIL combined DDP groups were 45.14%,57.98% and 77.23% .(2) Compared with single drug group,TRAIL combined with DDP could increase the apoptotic rate of liver cancer cells obviously.Conclusions Combined application of TRAIL and DDP has a synergistic inhibition to the transplanted HCC in mice.
