脂肪源性干细胞移植对脑缺血/再灌注损伤大鼠趋化因子及其受体表达影响的实验研究

摘    要:目的探讨脂肪源性干细胞(ADSCs)移植对大鼠脑缺血/再灌注损伤梗死区CXC趋化因子1(CXCL1)和CXC趋化因子2(CXCL2)及其受体CXCR2表达的影响。方法 24只实验动物随机分为假手术组、模型组和脂肪源性干细胞(ADSCs)组。ADSC组尾静脉注射PKH26标记的ADSCs细胞悬液,缺血72 h后行神经功能损害评分(NSS),免疫蛋白印迹法检测梗死区CXCL1、CXCL2和CXCR2蛋白表达。结果 ADSCs移植后可见PKH26标记的ADSCs在梗死区表达。与模型组比较,ADSC组NSS评分降低(P<0.05),梗死区CXCL1、CXCL2和CXCR2蛋白表达显著下降(P<0.05)。结论 ADSCs移植具有促进脑缺血损伤神经功能恢复的作用,其机制可能与其下调炎症趋化因子CXCL1和CXCL2及其受体CXCR2表达,抑制炎性反应有关。

Abstract:Objective To investigate the effects of transplantation of adipose-derived stem cell(ADSCs)on expressions of chemotatic factors CXCL1,CXCL2 and CXCR2 in infarction region after cerebral ischemia-reperfusion in rats.Methods Rats were randomly divided into sham operation group,model group and ADSCs group.ADSCs suspension were marked with PKH-26 and infused into the caudal vein in the ADSCs group.72h after ischemia,the neurological function was investigated by NSS method,and the expressions of CXCL1,CXCL2 and CXCR2 in the infarction area were detected by Western blot.Results PKH-26 labeled ADSCs were observed in infarction area.Compared with the model group,the NSS of ADSCs group was significantly decreased(P<0.05),and the expressions of CXCL1,CXCL2 and CXCR2 of infarction area were also significantly decreased(P<0.05).Conclusion ADSCs transplantation can promote cerebral ischemia injury neurological recovery,which may be related to the down-regulating of the expressions of infarction area CXCL1,CXCL2 and CXCR2,and restraining of the inflammatory response.
