孕期染毒PFOS对雄性胎鼠生殖系统的影响

摘    要:目的评价孕鼠染毒全氟辛烷磺酸盐(PFOS)对胚鼠生殖系统的影响。方法取孕鼠16只随机分为4组:0.05%TW-20(对照组)、5 mg/(kg.d)低剂量灌胃组、10 mg/(kg.d)中剂量灌胃组、20 mg/(kg.d)高剂量灌胃组,各4只。SD大鼠从怀孕第11天开始灌胃给予PFOS,第19天结束灌胃,第20天处死孕鼠取组织。观察测量平均妊娠胎鼠数、胎鼠雌雄比例和雄性胎鼠体质量、睾丸重量,采用real-time聚合酶链反应法测定睾丸Cyp17α1的相对表达量。结果四组孕鼠平均妊娠胎鼠数、雄性胎鼠体质量、睾丸重量比较差异有统计学意义(P<0.05);与正常对照组比较,各给药组睾丸内Cyp17α1 mRNA表达明显下降(P<0.01)。结论孕期染毒PFOS对胚胎发育和雄性生殖系统有毒性作用,不同剂量具有不同的效应。

Abstract:Objective To investigate the influence on reproductive of male fetus rats exposed to perfluorooctane sulfonate(PFOS).Methods 16 SD mice were divided into 4 groups randomly.The pregnant rats were exposed to 0.05% TW-20(control group)and PFOS at dose of 5 mg/(kg·d)(low-dose group),10 mg/(kg·d)(medium-dose group)and 20 mg/(kg·d)(high-dose group)by gavage from 11th day to 19th day of gestation.The pregnant rats were killed on the 20th day and the testes of male fetus rats were taken out after the exposure.The number of the fetus,the percentage of the male fetus,body weight and testes weight were measured after the exposure.The mRNA of Cyp17α1 were detected quantitative by real-time-PCR.Results Differences in pregnant rats average pregnancy fetal rat,male fetal rat body weight,testicular weight of the four groups were statistically significant(P<0.05).Compared with the normal control group,Cyp17α1 mRNA testicular expression of each group was decreased significantly(P<0.01).Conclusion Exposure to PFOS has toxic effect to embryo development and male reproductive system,which present different effects at different dosages.
