IgE高亲和力受体MS4A2基因单核苷酸多态性与哮喘气虚体质患儿相关性研究

摘    要:目的探讨IgE高亲和力受体MS4A2基因外显子及其侧翼区碱基序列单核苷酸多态性与哮喘气虚体质患儿相关性。方法选取58例气虚体质哮喘患儿作为哮喘组,50例健康体检儿童作为实验对照组。提取哮喘组和对照组外周血基因组DNA样本,应用PCR技术扩增MS4A2基因外显子及其侧翼区序列。扩增产物直接测序,确定碱基类型。结果通过测序结果分析,发现三个内含子多态(rsl441586,rs2847663,rs512555),一个外显子多态(rs569108)。rs569108位点AG基因型频率高于正常对照组(X2=7.938,P=0.0189)。结论rs569108位点AG型多态可能是导致气虚体质患儿哮喘的易感因素。

Abstract:Objective- To investigate the association of the qi deficiency asthma patients and the polymorphism of the MS4A2 gene exons and their flanking sequences. Methods: 58 cases of qi deficiency asthma patients and 50 health control were enrolled the study. Genomie DNA was extracted from peripheral blood leukocytes. The MS4A2 gene ten exons and their flanking sequences were amplified with polymerase chain reaction, and then subjected to direct DNA sequencing. Results: According to the DNA sequencing analysis, three intron polymorphism (rs1441586, rs2847663, rs512555) and one exon polymorphism (rs569108) were found. The rs569108 AG genetype frequency was higher in qi deficiency asthma patients than that in healthy cotrol (X2=7.938, P= 0.0189) . Conclusion: The AG genetype of rs569108 may be the suceptibility factor to qi deficiency asthma patients.
