白藜芦醇改善高脂饮食小鼠的糖代谢并棕化腹股沟白色脂肪

摘    要:目的:观察白藜芦醇治疗对高脂饮食诱导肥胖小鼠的体重,体脂比及血糖代谢的作用,初步探索白藜芦醇对白色脂肪棕化的作用。方法:小鼠高脂饮食(45%热量来自脂肪)8周后随机分为对照组(HFD—Con)和白藜芦醇治疗组(HFD—Res,白藜芦醇400mg/kg),治疗2周后称重、测定糖耐量和计算体脂比例,实时PCR测定腹股沟皮下白色脂肪组织解耦联蛋白-1(UCP-1)和cIDEA基因表达。结果:与HFD.Con组相比,HFD—Res组体重下降明显,糖耐量改善,脂肪组织/体重比无改变;而腹股沟皮下白色脂肪组织解耦联蛋白-1(UCP-1)和CIDEA基因表达升高。结论:白藜芦醇治疗可降低高脂饮食诱导肥胖小鼠体重和改善糖代谢,同时可能促进腹股沟皮下白色脂肪组织棕化。

Abstract:Objective: To investigate the effects ofresveratrol on the body weight, ratio of white adipose tissue, glucose level and UCP-1, CIDEA mRNA expression in high fat diet-induced obese mice. Methods: Obese mice induced by 8 weeks' high fat diet (HFD) were treated with resveratrol at a dose of 400 mg/kg (HFD-Res) or placebo (HFD-Con) for 2 weeks and were weighted. The ratio of white adipose tissue was calculated and glucose metabolism was evaluated by intraperitoneal glucose tolerance test (ipGTT). The gene expression of UCP-1 and CIDEA was determined by real-time PCR. Results: Compared with HFD-Con, HFD-Res had more weight loss (P〈0.05) while ipGTT was improved after 2 weeks' therapy. RT-PCR results showed that the expression of UCP-1 and CIDEA mRNA in the inguinal white adipose tissue was higher in HFD-Res than in HFD-Con (P〈0.05). Conclusion: Resveratrol has anti-obesity effect and can improve glucose metabolism in high fat diet-induced obese mice. Resveratrol can increase the gene expression of UCP-1 and CIDEA in inguinal white adipose tissue, indicating its potential effect in browning of white adipose tissue.
