| 摘 要： | 目的采用高效液相色谱-荧光法测定大鼠血液中睾酮的含量。方法色谱柱为Alltech C18柱(150 mm×4.6 mm,5μm),流动相为甲醇-乙醇(90∶10),荧光检测器,检测波长为λex=360 nm和λem=420 nm。结果血睾酮质量浓度在108～1 620 ng/mL范围内与峰面积线性关系良好,最低检出限为10.8 ng。高、中、低3个质量浓度平均回收率分别为93.14%(RSD=3.00%),85.99%(RSD=9.90%),80.14%(RSD=4.04%)。日内精密度的RSD为4.34%。结论高效液相色谱-荧光法可用于血液中血睾酮的含量测定。

| Abstract: | Objective To establish an HPLC- fluorescent method for determining the serum content of methyhestosterone. Methods The chromatographic conditions were the Alltech C18 column（150 mm ×4.6 mm, 5 μm）, methanol-ethanol （90：10） as the mobile phase, the emanation detection wavelength at 420 nm and absorption detection wavelength at 360 nm. Results The linear range of methyltestosterone mass concentration was within 108- 1 620 ng/mL. The lowest detection limit was 10. 8 ng. The average recovery rates of high, middle and low concentrations were 93.14% （ RSD = 3.00% ）, 85.99% （ RSD = 9. 90% ）, 80. 14% （ RSD = 4.04% ）, respectively. The in- traday precision RSD was 4. 34%. Conclusion The established HPLC-fluorescent method is suitable for the determination of serum methyltestosterone. |
