Abstract:Objective:To establish HPLC-ELSD fingerprint combined with statistics method for researching Kulianpi(Meliae Cortex)from different habitats.Methods:HPLC-ELSD method was used to establish the fingerprints of Kulianpi(Meliae Cortex),and the similarity evaluation,cluster analysis and principal component analysis were used to study the HPLC-ELSD fingerprints of 12 batches of Kulianpi(Meliae Cortex),and the content of toosendanin was determined.HPLC-ELSD analysis was performed on Thermo Acclaim C18(4.6 mm×250 mm,5μm),with a mobile phase consisting of acetonitrile-0.1%methanoic acid at the flow rate of 1.0 mL/min,and the column temperature was controlled at 30℃,the parameters of the evaporative photodetector were a drift tube temperature of 110℃and gas flow rate of 2.7 L/min.Results:HPLC-ELSD fingerprint of Kulianpi(Meliae Cortex)was established,and 8 common peaks were calibrated.The similarity of 12 batches of samples ranged from0.720 to 0.998.The samples were roughly divided into two categories by cluster analysis and principal component analysis.The samples from Guangdong and Guangxi were classified into one category,and the samples from Anhui and Hubei were classified into the other category.The content determination results showed that the linear relationship of toosendanin was good in the range of 19.682-984.116μg,and the average recovery rate was 103.65%.According to the comprehensive score,the quality of Kulianpi(Meliae Cortex)from Anhui and Hubei was better;according to the content of toosendanin,the quality of Kulianpi(Meliae Cortex)from Guangdong and Guangxi was better.Conclusion:The established fingerprinting method of Kulianpi(Meliae Cortex)is accurate and reliable with good repeatability,which can provide a reference for the quality evaluation of Kulianpi(Meliae Cortex).
