柘树根多糖CPS-1的分离纯化及理化性质分析

摘    要: 目的从柘树根中分离纯化出柘树根多糖CPS-1,并研究其理化性质。方法用水浸提柘树根,过3520树脂柱,乙醇沉淀,Sevag法脱蛋白,透析后经DEAE-Sephadex A-50柱纯化得CPS-1。用HPLC,TLC,GC,元素分析,IR,1H-NMR及13C-NMR研究CPS-1的理化性质。结果CPS-1达到高效液相色谱纯,由鼠李糖、阿拉伯糖、木糖、甘露糖、葡萄糖和半乳糖组成,物质的量之比依次为1.00∶14.02∶0.74∶2.62∶30.06∶17.72,CPS-1的糖苷键为α-构型,具有吡喃型糖环,C-2,C-3,C-4中某一碳上发生取代,C-6未发生取代。结论CPS-1是由鼠李糖、阿拉伯糖、木糖、甘露糖、葡萄糖和半乳糖组成的杂多糖,其数均相对分子质量(Mn)为4461,重均相对分子质量(Mw)为6572。

Abstract:OBJECTIVE To purify the polysaccaride from the root of Cudrania triouspidata (Carr.)Bur and study its physical and chemical properties. METHODS CPS-1 polysaccharide was isolated and purified from the root of Cudrania triouspidata (Carr.) Bur by methods of hot water extraction, 3520 resin column chromatography, ethanol precipitation, deproteination with Sevag method and DEAE-Sephadex A-50 column chromatography. Its physical and chemical properties was analyzed by HPLC, TLC, GC, element analysis,IR, 1H-NMR and 13C-NMR. RESULTS CPS-1 was showed to be homogeneous in HPLC. It was composed of Rha,Ara,Xyl,Man,Glu and Gal in a molar ratio of 1.00∶ 14.02∶0.74 ∶2.62∶30.06∶17.72 in turn. The glycoside bond configuration of CPS-1 was α. Its monosaccharide contained pyranose structure and the substitution was located on the position of C-2 or C-3 or C-4. The substitution was not located on the position of C-6. CONCLUSION CPS-1 is a kind of heteropolysaccharide composed of Rha,Ara,Xyl,Mal,Glc and Gal. Its number-average molecular weight is 4 461 and the weight-average molecular weight is 6 572.
