毛茛有效部位D1对人胃癌细胞MGC803血管生成拟态的作用及其机制

摘    要:目的以毛茛有效部位D1为研究对象,研究其对人胃癌细胞MGC803血管生成拟态的作用。方法采用细胞粘附实验检测毛茛有效部位D1对人胃癌细胞MGC803粘附作用的影响;采用细胞划痕实验观察D1对MGC803细胞迁移能力的影响;采用Matrigel肿瘤细胞类血管生成模型观察D1对MGC803体外类血管生成的影响;采用半定量RT-PCR方法检测D1对MGC803血管生成拟态相关基因的作用。结果毛茛有效部位D1可有效地抑制人胃癌细胞MGC803细胞粘附、细胞迁移和人胃癌细胞MGC803介导的血管生成拟态并呈剂量依赖性。半定量RT-PCR实验结果显示,D1能明显抑制MGC803血管生成拟态相关基因Sema 4D、VE-Cadherin、MMP2和Integrinβ5的表达。结论毛茛有效部位D1通过抑制血管生成拟态相关基因Sema 4D、VE-Cadher-in、MMP2和Integrinβ5的表达来抑制人胃癌细胞MGC803细胞粘附、细胞迁移和血管生成拟态,提示毛茛有效部位D1是一个抗胃癌血管生成拟态的候选药物。

Abstract:Aim To study the effect of ranunculus japonicus components D1 on vasculogenic mimicry of human gastric cancer cell MGC803 in vitro and its possible mechanisms.Methods The effect of D1 on the cell adhesion of human gastric cancer cell MGC803 was observed by cell attachment assay.The effect of D1 on the cell migration of MGC803 was observed by cell wound healing assay.The function of D1 on the tube-like structures formation in human gastric cancer cell MGC803 in vitro were observed by tube formation assay.The effect of D1 on the expression of vasculogenic mimicry related genes was assessed by semi-quantitative RT-PCR assay.Results Ranunculus japonicus components D1 effectively inhibited human gastric cancer cell MGC803 cell adhesion,cell migration and cancer cell mediated vasculogenic mimicry in vitro in a dosage-dependent manner.Furthermore,the semi-quantitative RT-PCR showed that D1 markedly inhibited the expression of vasculogenic mimicry related genes VE-Cadherin,Sema 4D,MMP2 and Integrin β5.Conclusion Ranunculus japonicus components D1 can effectively inhibit human gastric cancer cell MGC803 cell attachment,cell migration and cancer cell mediated vasculogenic mimicry by down regulating vasculogenic mimicry related genes VE-Cadherin,Sema 4D,MMP2 and Integrin β5.The study suggests that D1 is a promising candidate of anti-gastric cancer mediated vasculogenic mimicry.
