激光氧液对癫痫持续状态大鼠学习记忆及海马神经元突触素表达的影响

摘    要:目的:观察激光氧液对戊四氮(PTZ)所致癫痫持续状态(SE)大鼠学习记忆功能及海马神经元突触素(P38)表达的影响.方法:28日龄雄性SD大鼠以PTZ建立癫痫持续状态模型,随机分为PTZ+激光氧液组、PTZ+GS组和NS+激光氧液组、NS+GS组,分别给予激光氧液和5%葡萄糖液(5%GS)尾静脉输入,于模型制作前及模型成功后第0 d、3 d、7 d、10 d、14 d通过Y型电迷宫检测大鼠学习记忆能力,并于模型成功后各时间点通过免疫组化方法观察海马区P38阳性细胞数.结果:①行为学结果:PTZ两组大鼠SE发作后0 d大鼠学会达标次数较相应的NS组明显增多(P<0.05).PTZ+激光氧液组3 d、7 d时大鼠学会达标次数较PTZ+GS组大鼠明显减少(P<0.05).各组大鼠经过多次电迷宫刺激后,随时间延长学会达标次数均逐渐减少;②PTZ+GS组大鼠P38阳性细胞数在SE后3 d时较NS+GS组明显减少(P<0.05),在7 d时增加到接近或超过NS+GS组水平,在10 d、14 d较NS+GS组明显增加(P<0.05).PTZ+激光氧液组P38阳性细胞数与PTZ+GS组相比在7 d、10 d、14 d逐渐增多(P<0.05).结论:激光氧液有助于改善SE大鼠学习记忆能力,增强SE后发育期大鼠海马P38的表达.

Effect of Laser Oxygen Liquid Extract on Learning and Memory and the Expression of P38 of Hippocampal Tissues for Status Epilepticus Rats

Abstract:Objective:To investigate the therapeutic mechanism of laser oxygen liquid on learning and memory ability and the expression of synap tophysin(P38) of hippocampal neurons of rats following pentylenetetrazol-induced status epilepticus.Methods:Postnatal day 28(P28) Sprague Dawley(SD) rats received pentetrazole(PTZ)intra-abdominaly to make the model of status epilepticus(SE).The rats were divided randomly into PTZ group、 PTZ+laser oxygenate fluid group、 normal saline(NS)+GS group and NS+laser oxygenate fluid group.Each group was injected laser oxygen liquid and glucose of 5% from tail vein.The abilities of learning and memory of rats were tested by Y-type maze separately before the rat models were made and after the rat models were succeeded for 0 day、3 day、7 day、10 day and 14 day After the rat models were succeeded for 0 day、3 day、7 day、10 day and 14 day,the expressions of P38 in hippocampal were watched by immunohistochemical method.Results:①The learn standard number of two PTZ groups with SE has an obvious increase comparing with the NS groups(P0.05) after 0 day.The learn standard number of rats in PTZ laser oxygen liquid group has decreased after 3 day and 7 day(P0.05).The difference of cognitive ability was not significant in two groups after 10 day and 14 day(P0.05).The learn standard number for four groups after 10 day trainings has decreased comparing with that after first training(P0.05);②The number of P38 positive cells in CA1 for the rats of PTZ+GS group with SE after 3 d has decreased obviously comparing to that of NS+GS group(P0.05).It increased nearly to the standard of NS+GS group after 7 d(P0.05) and increased obviously after 10 d、14 d(P0.05).The number of P38 positive cells for the rats of PTZ laser oxygen liquid group after 7 d 、10 d、14 d has increased comparing to that of PTZ+GS group(P0.05).The change in CA3 has a similar tendency,but the number of P38 positive cells for the rats of PTZ+GS group after 7 d has increased comparing to that of NS+GS group(P0.05).Conclusions:The learning and memory ability could be improved and the expression of P38 in hippocampal tissues of the SE rats has been increased by laser oxygen liquid.
