氯化镉对小鼠脾细胞增殖与细胞钙稳态关系的影响

摘    要:以小鼠脾淋巴细胞转化,白介素2(IL-2)产生,胞内游离钙浓度([Ca2+]i),钙调蛋白量为指标,观察氯化镉(CdCl2,10-200μmolL-1)对脾细胞增殖的影响与细胞钙稳态的关系.结果表明,在本实验条件下,CdCl2对小鼠脾淋巴细胞转化,IL-2的产生有明显抑制作用,且与剂量和时间呈依赖关系.CdCl2对脾细胞钙稳态有干扰,致细胞[Ca2+]i浓度升高,钙调蛋白量下降,也与剂量和时间呈依赖关系.提示CdCl2对小鼠脾细胞增殖的抑制作用与细胞钙稳态失衡密切相关.

Abstract:In this study, mouse splenocytes were used to determine the effects of CdCl2(10-200 μmol·L-1) on lymphocyte transformation, interleukin 2 (IL-2) production, intracellular free Ca2+ concentration ([Ca2+i), and calmodulin activity. The results showed that CdCl2 inhibited lymphocyte transformation and IL-2 production with a significant linear dose and time response relationship. CdCl2 also significantly enhanced mouse splenocyte [Ca2+i and reduced calmodulin activity with a linear dose and time-response relationship. This study demonstrated a strong association between the inhibition of splenocyte proliferation and changes in calcium homeostasis caused by Cd.
