肿瘤坏死因子α对妊娠早期人滋养层细胞分泌绒毛膜促性腺激素的调节机理研究

摘    要:从正常妊娠早期人胎盘中分离出滋养层细胞,经重组肿瘤坏死因子α(rTNF-α)刺激,MTT生物检测法测定其上清液白细胞介素6(IL-6)的活性,同时用酶免疫分析法检测人绒毛膜促性腺激素(hCG)的活性。结果显示,被rTNF-α刺激之后,合体滋养层细胞分泌IL-6先于hCG.当用抗IL-6受体的单克隆抗体PM-1阻断后,hCG分泌量明显下降,而此时IL-6的产生量不受影响。当滋养层细胞经rTNF-α刺激产生IL-6之后再用PM-1阻断IL-6受体,则不影响hCG的分泌。结果表明,肿瘤坏死因子刺激合体滋养层细胞先产生IL-6,然后再通过其IL-6受体信号系统调节hCG的分泌。

Abstract:bjective:To investigate the regulatory effect of tumornecrosis factor-α(TNF -α)on th production of hCG by huma trophoblast cells . Design :An in vitro experimental study . etting:Laboratory in Osaka University Medical School . Material: Trophoblast cells were isolated from human placenta of early pregnancy Recombinant TNF-α,IL-1,IL-6 and IL-6 receptor monoclonal antibody PM-1 were obtained from Japan . Methods: Trophoblast cclls were isolated from hunad placenta of early pregnancy with percoll methodand incubated with rTNF-α hCG concentration in the medium was measured with EIA, and IL-6 activity was measured by MTT method. Results:,stimu1ation with rTNF-α the trophot cells secreted IL-6 earlier than hCG.When blocked with the monoclonal antibody of IL-6 receptor ,PM-1, hCG secretion significantly decreased,but the IL-6 production is not affected. If rTNF-α stimulated the production of IL-6 befre the use of PM-1 to block the IL-6 receptor ,then the secretion of hCG was not affected.concl usion :The results indicated that rTNF -α stimulated the trophoblast cells to produce IL-6,and through the Il-6 receptor system regulate the hCG secretion.
