黑面神枝叶水提物抑菌有效部位的筛选研究

摘    要:目的:筛选黑面神枝叶水提物的抑菌有效部位。方法:运用系统溶剂萃取法将黑面神枝叶水提物分为氯仿、乙酸乙酯、水饱和的正丁醇及剩余水层部位,采用试管二倍稀释法分别进行4个不同极性部位对金黄色葡萄球菌、耐药金黄色葡萄球菌及铜绿假单胞菌的体外抑菌实验研究,用以筛选抑菌作用的有效部位,并测定对敏感菌的最低抑菌浓度(MIC)和最低杀菌浓度(MBC)。结果:乙酸乙酯部位对3种菌种的抑菌作用最强,其MIC均为62.5 mg/m L,MBC均为250 mg/m L;其次为水饱和的正丁醇部位和剩余水层部位,其对试验菌种的MIC分别为125 mg/m L和62.5 mg/m L,正丁醇部位的MBC分别为500 mg/m L、500 mg/m L、250 mg/m L;氯仿部位对试验菌种均无抑菌作用。结论:黑面神枝叶水提物抑菌有效成分主要集中于乙酸乙酯部位和正丁醇部位。

Abstract:Objective: To preliminarily study the antibacterial effective parts of water extracts of branch- leaves from Breynia fruticosa. Methods: Using fluorescence methods,the water extracts of branch- leaves from Breynia fruticosa were divided into four parts,including chloroform,ethyl acetate,N- butyl alcohol and remaining water part. Using liquid media two- fold dilution method,bacteriostasis experiment studies in vitro are made respectively to staphylococcus aureus,resistant staphylococcus aureus and pseudomonas aeruginosa of four different polar parts to screen effective parts of bacteriostasis,and determine the sensitive bacteria minimum inhibitory concentration( MIC) and minimum bactericidal concentration( MBC). Results: Ethyl acetate part showed the strongest bacteriostasis to three bacteria. All the MIC was 62. 5 mg/m L,MBC was 250 mg / m L,followed by the N- butyl alcohol of water saturation part and remaining water part. The MIC of the tested strains was 125 mg / m L and 62. 5 mg / m L and the MBC of N- butanol part was 500 mg / m L,500 mg / m L and 250 mg / m L. Chloroform part had no bacteriostasis to the test strains. Conclusion: The antibacterial active ingredients of water extracts of branch- leaves from Breynia fruticosa are mainly concentrated in ethyl acetate part and N- butanol part.
