摘    要:目的:探讨骨髓间充质干细胞(BMSCs)在体外对人外周血T淋巴细胞的免疫调节作用。方法:从人骨髓中分离培养BMSCs,采用流式细胞仪(FCM)分析鉴定BMSCs的纯度。在PHA刺激下,人外周血淋巴细胞与不同数量的BMSCs共培养。检测各组T淋巴细胞的增殖、凋亡情况,分析各组T淋巴细胞免疫表型CD152、CD28的表达以及各组T淋巴细胞的IFN-γ、IL-10、TGF-β1 mRNA的表达。结果:在体外,BMSCs对由PHA诱导的T淋巴细胞的增殖、凋亡均有抑制作用,且抑制作用与BMSCs呈剂量依赖性。BMSCs对T淋巴细胞的CD152、CD28的表达无明显影响。BMSCs能促进IL-10、TGF-β1mRNA的表达,抑制IFN-γmRNA的表达。结论:BMSCs可能通过抑制T淋巴细胞增殖、减少T淋巴细胞凋亡、促进T淋巴细胞IL-10、TGF-β1mRNA表达及抑制IFN-γmRNA表达来下调免疫反应。

Abstract:Objective:To investigate the immunoregulatory effects of Bone mesenchymal stem cells(BMSCs)on human peripheral blood T-lymphocytes in vitro.Methods:BMSCs were isolated and expanded from bone marrow cells,The purity of BMSCs was identified by flow cytometry(FCM).The BMSCs(4×104、1×104、2×103)were added into wells containing lymphocytes(2×105)from unrelated donors in the presence of phytohemagglutinin(PHA).We assessed the proliferation of T-lymphocytes,the immunophenotypes(CD152,CD28)of T-lymphocytes were analyzed by flow cytometry.We also observed the apoptosis of T-lymphocyte of and the mRNA expression of interferon γ(IFN-γ),interleukin-10(IL-10),transforming growth factor-β1(TGF-β1).Results:In all the experiments,with the presence of BMSCs,human peripheral T-lymphocytes showed a statistically significant decrease of their proliferative activities and apoptosis in a dose-dependent manner.No inhibitory effects on CD152and CD28 of T-lymphocytes had been observed.BMSCs promoted the mRNA expression of IL-10,TGF-β1 and inhibited the mRNA expression of IFN-γ.Conclusion:BMSCs can inhibit the proliferation of human peripheral T-lymphocytes and up-regulate the expression of IL-10,TGF-β1 and down-regulate the expression of IFN-γ,which may play an important role in it's immunoimpressive affects.
