摘    要:目的探讨鲑鱼鱼白DNA(Salmon milt DNA,SMD)对小鼠胸腺增龄性萎缩作用的分子机理。方法10月龄雌性Balb/c小鼠按体质量随机分为实验组和对照组,在常规饮食的基础上各组每日分别灌胃333.33和0mg/kg的SMD,5周后,无菌取胸腺;应用RT-PCR和Northern blot技术检测Genebank登录号为Aw209102、U23789、X80232、NM_009678和AF083929的基因在对照组和实验组胸腺组织中的表达情况。结果Aw209102、U23789、X80232在实验组表达上调;NM_009678、AF083929在实验组表达下调。结论SMD可通过促进细胞增殖相关基因和发育、分化相关基因表达,并抑制细胞凋亡相关基因的表达而逆转胸腺增龄性萎缩。

Abstract:Objective To investigate the molecular mechanism of salmon milt DNA (SMD) in age-related involution of mouse thymus. Methods Ten-month-old female Balb/c mice were divided randomly into experiment and control groups according to their weights. The mice in the experiment group were supplied with SMD (333.33 mg/kg) but the mice in the control group were not supplied with SMD. RT-PCR and Northern blot were used to detect the expressions of Aw209102, U23789, X80232, NM009678, and AF083929 genes in all their thymuses. Results The expression levels of Aw209102, U23789, and X80232 genes were higher in experiment group those in control group, but the expression levels of NM009678 and AF083929 genes were lower in experiment group than in control group. Conclusion SMD may reverse the age-related involutions in mouse thymus via up-regulating the expression of proliferation related genes as well as the expression of development and differentiation related genes, and via down-regulating the expression of apoptosis related genes simultaneously.
