秀山金银花、叶、茎中绿原酸的检测及其抑菌作用研究

摘    要:目的测定金银花及其叶片、茎中绿原酸的含量,考察其抑菌效果。方法采用高效液相色谱法(HPLC法)和最低抑菌浓度(MIC)测定法。色谱柱为Phenomenex C18柱(250mm×4.6mm,5μm),流动相为乙腈-pH=2.04的0.4%磷酸(13:87,V/V),流速为0.5mL/min,检测波长为327nm。结果金银花及其叶片、茎中绿原酸的含量分别为6.1%,1.8%,0o金银花提取液和金银花叶片提取液对大肠杆菌的MIC分别为18.75mg/mL和150mg/mL,对金黄色葡萄球菌的MIC分别为4.6875mg/mL和37.5mg/mL。结论金银花及其叶片中绿原酸含量高,抑菌作用强。仝银花叶片具有较高的开发价值,药效可代替金银花。

Abstract:Objective To detect the amount of chlorogenie acid in bud, leaf and stem of flos lonicerae respectively and to examine the effects of its antibacterial activities, Methods High performance liquid chromatography (HPLC) was used to determine the amount of chlorogenic acid and the antibacterial effect was expressed by its minimal inhibitory concentration (MIC). The column was Phenomenex C18 chromatography column (250 mm× 4. 6 mm, 5 μm); the mobile phase was acetonitrile -0. 4%- phosphonic acid (13 : 87, V/ V), pH = 2.04; the flow rate was 0.5 mL/min; A = 327 nm. Results The contents of chlorogenic acid in flos lonicerae bud and its leaf, stem were 6.1%, 1.8% and 0 respectively. The MIC of extracts from flos lonicerae and its leaf against eschcrichia coil were 18.75 mg/mL and 150 mg/mL respectively; the MIC against staphylococcus aureus were 4. 687 5 mg/mL and 37.5 mg/mL respectively. Conclusion Leaf of flos lonicerae has strong exploiting value, and the drug effect of flos lonicerae can be replaced by chlorogenic acid extracted from its leaf.
