血竭素高氯酸盐促进瘢痕成纤维细胞凋亡的作用

摘    要:目的观察天然药物血竭素高氯酸盐对瘢痕成纤维细胞凋亡的影响,为开发有效治疗增生性瘢痕的新药提供实验依据。方法应用四甲基偶氮唑(MTT)法、特异性荧光染色和原位末端标记技术(TUNEL)分别检测血竭素高氯酸盐对成纤维细胞的抑制率和凋亡指数。结果MTT法显示血竭素高氯酸盐对成纤维细胞的抑制率增高,且呈现浓度和时间依赖性(P<0.05)。特异性荧光染色和TUNEL法检测凋亡指数,均呈剂量依赖性增高(P<0.05)。结论血竭素高氯酸盐对瘢痕成纤维细胞的增殖具有显著的抑制作用,且呈量效关系促进成纤维细胞发生凋亡。

Abstract:Objective To observe the effect of herbal medicine dracorhodin perchlorate(Dp)on apotosis in scar-derived fibroblasts and offer experimental evidence for new drug of treating human hyperplastic scars.Methods The fibroblasts were cultured as an experimental model and the effect of Dp were detected.The cell proliferation was determined by MTT assay,the inhibiting effect fluorescent staining and TUNEL analyzed apoptosis of fibroblasts induced by Dp.Results It was shown that exposure to Dp could increase the inhibiting rate of fibroblasts by MTT assay and dependent on concentration and time(P<0.05),otherwise it could significantly elicit the apoptosis of cultured fibroblasts by fluorescent staining and counting TUNEL-positive cells (P<0.05).Conclusion Dp could significantly inhibit the proliferation and accelerate the apoptosis of hyperplastic scar fibroblasts in vitro in a dose-dependent relationship.
