水囊压迫大鼠脑组织致星形胶质细胞中胶质纤维酸性蛋白的变化

摘    要:目的研究水囊压迫大鼠颅脑造成损伤后星形胶质细胞的反应。方法将改装后的5F漂浮导管球囊部位全部插入颅内硬膜外后,注射0.5ml无菌生理盐水人球囊使其扩张并维持压力2h。于伤后不同时间将大鼠处死,取脑组织进行HE染色和胶质纤维酸性蛋白(Glialfibrillaryacidicprotein,GFAP)免疫组化染色。结果伤后存活1h组,可见GFAP阳性细胞散在于损伤区周围,但突起较短,反应性星形胶质细胞主要局限于创缘周围。伤后存活24h组,GFAP表达的范围扩大,在创缘及其伤区周围的阳性细胞增多,胞体增大,突起增长,且染色加深。24h在海马组织中已有GFAP阳性细胞表达。伤后存活3d、6d和12d组,阳性细胞的表达继续维持在高水平,且细胞形态各异,胞体变化较大,在创缘出现突起少而粗短的GFAP阳性细胞,而在损伤区周围、海马以及脑干的区域阳性细胞突起粗大不规则,突起进一步增长,胞浆突起染色较深。12d时,GFAP阳性细胞增多达到高峰,阳性细胞着色深,呈深棕黄色。结论水囊压迫造成脑组织损伤后星形胶质细胞的标志性蛋白GFAP表达出现规律性变化,表明水囊压迫的损伤模型可以模拟颅脑血肿的损伤。

Abstract:Objective To investigate the response of astrocytes to craniocerebral injury caused by water-sac pressure on rat brain.
