环氧化酶-2在肾缺血再灌注损伤的实验研究

摘    要:目的检测大鼠肾缺血再灌注(I/R)损伤90分钟后COX-2的表达,探讨两者的关系。方法构建I/R模型,取30只大鼠平均分为6组,分别于再灌注后0、1.5、3、5、12和24 h处死,采用免疫组化染色分析分为0-4等级,计算COX-2的表达。结果COX-2内皮细胞的表达在再灌注后3~5 h最明显,12~24 h后逐渐减弱。COX-2的表达在再灌注后1.5、3、5、12和24 h均明显高于0 h(P<0.05)。COX-2表达的最高峰后几小时可发现肾(I/R)损伤最明显。结论COX-2表达和肾(I/R)损伤有关。

Abstract:Objective To explore the expression of cyclooxygenase 2(COX-2) in rats' kidneys after 90 minutes of ischemia-reperfusion(I/R)injury,and study the relation of COX-2 and renal I/R injury.Methods 30 Rats I/R model were randomly divided to 6 groups and sacrificed at 0,1.5,3,5,12,and 24hours after reperfusion.COX-2 expression was detected by immunohistochemical staining,which was graded 0 to 4.Results COX-2 expression was most intense on endothelial cells at 3 and 5 hours after reperfusion and gradually became weaker from 12 to 24 hours after reperfusion.The COX-2 expression index were significantly higher at 1.5,3,5,12,and 24 hours after reperfusion than that at 0 hour(P<0.05).Several hours after the maximum of COX-2 expression the maximum renal I/R injury was observed.Conclusion There was a relationship between COX-2 expression and renal I/R injury.
