一点红中总生物碱的提取与测定方法研究

目的：探讨超声波法和索式提取法对一点红中总生物碱的最佳工艺条件与测定方法的研究。方法：以紫外分光光度法为测定方法，一点红中总生物碱提取来量为考察目标，采用正交实验法优化提取工艺。结果：不同温度、不同浓度、提取时间对一点红总生物碱提取量有显著影响。提取次数影响不大。结论：超声波法的提取最佳工艺为80℃下以物料比为1∶40对一点红生物碱类物质进行30min提取，得出提取率为0.435%，索式提取法的提取最佳工艺为用80%的乙醇对一点红生物碱类物质进行2h提取，得出提取率为0.320%。通过两种方法的比较，一点红总生物碱的提取为超声波法更优。

Extraction and Determination of Total Alkaloid about Emilia Sonehifolia

Objective:discussing the optimum extraction conditions and determination method of total alkaloid in Emilia Sonehifolia using the method of ultrasonic extraction and soxhlet extraction. Methods:use the method of UV spectrophotometry to detect the content of total alkaloid, to optimize the extraction process by orthogonal experiment method. Results: extraction quantity of total alkaloid has significant effect in the different temperature, ethanol concentration and extracting time. Conclusion:the best process of extraction was using 1:40 of material ratio for 30 min under the 80℃in ultrasonic method, and extraction rate was 0.435%. the best process of extraction in soxhlet? extraction as follows: the duration of extraction was 2h with 80% ethanol, and extraction rate was 0.320%. Through the comparison of two methods, we concluded that the best method of extraction of total alkaloids in Emilia sonehifolia is ultrasonic method.
