实验性脑出血大鼠血肿周围补体C3表达的动态观察

摘    要:目的 探讨补体激活在脑出血后脑水肿形成中的作用。方法 雄性SD大鼠48只,随机分为假手术组和脑出血组。立体定向注血制作脑出血动物模型,在手术后6h、12h、24h、48h、72h、7d分别处死动物,干湿重法测量脑含水量,免疫组化法测量血肿周围补体C3阳性细胞数,观察血肿周围炎细胞浸润情况。结果 血肿周围补体C3表达于6h开始增多,72h达到高峰,7d仍高于假手术组,血肿周围炎细胞浸润和脑水肿也于出血后48—72h达到高峰,提示补体C3表达与炎细胞浸润及脑水肿有关。结论 补体C3参与了脑出血后脑水肿的形成。

Abstract:Objective To investigate the activation and effects of complement 3 in brain edema formation after intracerebral hemorrhage (ICH).Methods Forty-eight healthy male Sprague-Dawley rats were divided randomly into sham-operation group and intracerebral hemorrhage group. Autologous artery blood was infused stereotactically into the right basal ganglia to establish the model. Rats were sacrificed separately at 6 h, 12 h, 24 h, 48 h, 72 h and 7 d to measure brain water content, to observe C 3 deposition and inflammatory cells infiltration in surrounding hematoma or within the clot by immunohistochemitry in each group.Results The expression of C 3 increased at 6 h and reached maximum at 72 h after blood injection, 7 d was still higher than that in sham-operation group. Inflammatory cells became apparent in perihematoma at 24 h, and were more abundant at 48 h, then decreased gradually till to disappear at 7 d. We demonstrated that C 3 deposition was significantly relative with inflammatory cells infiltration and brain edema.Conclusion Activation of complement 3 might play an important role in the formation of brain edema after ICH.
