汉黄芩素对胶质瘤U251细胞Survivin mRNA及Caspase-3mRNA表达的影响研究

摘    要:目的 探讨汉黄芩素对胶质瘤U251细胞Survivin mRNA及Caspase-3 mRNA表达的影响.方法 192株人胶质瘤U251细胞株分别用汉黄芩素(96株)、替莫唑胺(TMZ)(48株)处理,留取48株作为空白对照组,半定量RT-PCR法检测Survivin mRNA与Caspase-3 mRNA表达强度,并在3组之间进行对比分析.结果 空白对照组Survivin mRNA表达强度为(0.773±0.176) μM,显著高于汉黄芩素组的(0.382±0.087) μM及TMZ组的(0.408±0.092) μM (P < 0.05);Caspase-3 mRNA表达强度为(0.097±0.043) μM,显著低于汉黄芩素组的(0.454±0.213) μM及TMZ组的(0.432±0.187) μM(P < 0.05),汉黄芩组与TMZ组之间比较差异无统计学意义(P > 0.05).结论 汉黄芩素可通过下调Survivin mRNA的表达并上调Caspase-3 mRNA的表达起到有效诱导人胶质瘤U251细胞凋亡的作用.

Abstract:Objective To discuss the effects of wogonin on the expression of Survivin mRNA and Caspase-3 mRNA in human giloma cell line U251. Methods One hundred and ninety-two human giloma cell line U251 were cultured with wogonin (96 strains) and TMZ (48 strains), and other 48 strains were as blank control group, Survivin mRNA and Cas- pase-3 mRNA expression level were detected by semi-quantitative RT-PCR, and then Survivin mRNA and Caspase- 3 mRNA expression level were contrasted between the three groups. Results Survivin mRNA expression level in the blank control group was (0.773±0.176) μM, significantly higher than (0.382±0.087) μM in the wogonin group and (0.408±0.092) μM in the TMZ group (P 〈 0.05), and Caspase-3 mRNA expression level was (0.097±0.043) μM, sig- nificantly lower than (0.454±0.213)μM in the wogonin group and (0.432±0.187) μM in the TMZ group (P 〈 0.05), but there was no significantly difference in Survivin mRNA and Caspase-3 mRNA expression level between wogonin group and TMZ group (P 〉 0.05). Conclusion Wogonin play a role in inducing apoptosis in glioma U251 cells through re- ducing the expression of Survivin mRNA and raising the expression of Caspase-3 protein expression.
