脑胶质瘤中Hint1基因甲基化状态与表达的研究

目的探讨脑胶质瘤中Hint1基因启动子区的甲基化与表达情况。方法采用MSP和Western-blit法检测Hint1基因在脑胶质瘤和正常脑组织中甲基化状况及对应表达情况。实时荧光定量PCR的方法检测Hint1 mRNA的表达情况。结果经检测正常脑组织和胶质瘤Hint1基因启动子区域呈非甲基化状态,在83例胶质瘤患者脑组织中Hint1 mRNA的平均表达量为0.5190±0.0280,高于在30例正常脑组织中的平均表达量0.2236±0.0300。Hint1 mRNA的表达量与患者年龄、性别等临床特征无明显相关性而与患者的临床分期密切相关（P〈0.01）。结论脑胶质瘤中Hint1基因启动子区域呈非甲基化状态,其表达程度与患者的临床分期密切相关。

Research of glioma Hint1 gene methylation status and expression

Objective To explore methylation and expression of promoter domain in Hint1 gene in patients with glioma. Methods Methylation and expression of promoter domain in Hint1 gene from brain tissue of normal people and patients with glioma were tested by MSP and Western-Blot. mRNA level of Hint1 gene were tested by RT-PCR. Results The promoter domain of Hint1 gene presented unmethylated state both in normal people and in in patients with glioma. mRNA expression of Hint1 in the brain tissue of 83 patients with glioma was 0.5190 ±0.0280, higher than the average expression in 30 cases of normal brain tissue which was 0.2236 ±0.0300 with statistically significant differences （P 0.01） existed. Hint1 mRNA expression level showed no significant correlation with age, gender or other clinical features but was closely related to clinical stage of the patient. Conclusion Promoter domain in Hint1 gene in patients with glioma presents unmethylated state and Hint1 gene expression is closely related to patient＇s clinical stage.
