摘    要:目的探讨慢性牙周炎(CP)对5/6肾切除大鼠肾功能的影响。方法将40只健康雄性SD大鼠随机分为4组,每组10只。A:对照组,B:慢性牙周炎组,C:慢性肾衰竭组,D:慢性牙周炎+慢性肾衰竭复合组。于8周末处死动物,检测牙周附着丧失(AL)、改良出血指数(mBI)、血清肌酐(SCr)水平。检测血清(ELISA法)及肾组织中(免疫组织化学法)高敏C反应蛋白(hs—CRP)的表达,进行统计学分析。结果肾功能显示,C、D组SCr明显高于A、B组,D组显著高于C组,B组肾功能无下降。B、D组出现显著牙周炎症,D组mBI、AL水平高于B组。B、C、D组血清hs—CRP水平较A组显著升高,D组hs-CRP较B组升高。hs-CRP在B、C、D组肾脏均有表达,D组hs-CRP表达高于B组。结论本模型可用于观察研究牙周炎与慢性肾衰竭的相关性,慢性牙周炎可促进肾功能恶化,可能通过炎性机制加重肾脏的损害。

Effects of chronic periodontitis on renal function in rat model of 5/6 nephrectomy

Abstract:Objective To investigate the possible correlation between chronic periodontitis(CP) and chronic renal failure(CRF) by establishing chronic periodontitis and chronic renal failure model in SD rats. Methods Forty health male SD rats were divided into four groups: A(control group), B(CP group) ,C(CRF group), D (CP + CRF group), 10 rats in each group. All rats were sacrificed after 8 weeks to detect periodontal index and levels of serum creatine(SCr). The expression of hs-CRP in serum and tissue was examined by ELISA and immunohistochemistry. The data were statistically analyzed. Results SCr levels in groups C and D were higher than in groups A and B(P〈0. 05), and those in group D were higher than in group C (P〈0. 05). There was significant difference in SCr levels between groups A and B (P〈0.05). Obvious inflammation of periodontal tissue was observed in groups B and D. mBI and AL in group D were higher than those in group B(P〈0. 05). AL in group C was higher than that in group A(P〈0. 05). The serum hs-CRP level in groups B,C and D were higher than those in group A(P〈0. 05) ,and hs-CRP level in group D was higher than in group B(P〈0. 05). The expression of hs-CRP in renal tissue was compatible with that in serum. Conclusions The model can be used to observe the correlation between chronic periodontitis and chronic renal failure. Chronic periodontitis can aggravate chronic renal failure probably through the inflammatory mechanism.
