摘    要:目的 本研究旨在对鼠伤寒沙门氏菌LT2及其实验室诱导喹诺酮耐药株的耐药机制进行特征描述.方法 使用浓度递增法进行实验室中鼠伤寒沙门氏菌LT2的环丙沙星耐药株的诱导,并通过λRed噬菌体重组系统和噬菌体P22构建缺失株,进行染色体介导的喹诺酮耐药机制研究.结果与结论 实验室诱导获得喹诺酮耐药菌株LT2-4,经分析发现,其拓扑异构酶编码基因gyrA出现突变位点(S83F).acrAB和tolC基因的缺失致使LT2-4对环丙沙星敏感,提示它们在喹诺酮耐药机制中发挥重要作用,并发现tolC在二者中可能占有主导地位.

Abstract:Objective To describe the characterization of quinolone-resistant mechanisms in Salmonella enterica Typhimurium(S.Typhimurium) LT2and its lab induced drug-resistant strain.Methods The drugresistant mutant was selected by exposing S.Typhimurium LT2 to increasing concentrations of ciprofloxacin in MH broth.The characterization of the quinolone-resistant mechanisms was described through the construction of isogenic mutants using phage λRed recombinase system and phage P22.Results and Conclusion The quinolone-resistant strain LT2-4was obtained by lab induction,and the strain showed the occurrence of the point mutation(S83F) of the topoisomerase coding gene gyrA.The deletion of gene acrABand gene tolClead to enhancing sensitivity to ciprofloxacin in LT2-4,suggesting the two genes played an important role in quinolone-resistant mechanisms;in addition,tolCmight be in a dominant position.
