摘    要:目的建立人血浆中水飞蓟宾浓度的测定法。方法采用高效液相色谱法,μBondapak(300mm×4.6m,5μm)色谱柱,及μBondapak预柱(10mm×4.6mm,5μm)为分析柱。乙晴、水和二氧六环(20:70:10)混合溶剂为流动相,流速为1.0mL/min,检测波长为288nm,柱温为40℃。结果水飞蓟宾在5.123~327.9μg/mL范围内线性良好(r=0.9998),方法回收率为93.2%~96.8%,日内及日间RSD<10%。结论方法简便快速、灵敏度高,适合于临床药学研究需要。

Abstract:Objective To establish a precise, simple and convenient method for the determination of sylybin concentration in human plasma. Methods HPLLC system was used u Bondapak (300 mm×4.6 um) and (10 mm×4.6 um) as the column, mixture of acetonitrile, water and dioxane as the mobile phase. The flow rate of 1.0 mL/min and column temperature of 40℃ was adopted and the detection wavelength of 288 nm was specified. Results The calibration curve was linear over the range from 5.123 to 327.9 ng/mL, with r=0.9998. The recovery was 93.2~96.8%. The RSD for within day variation and the between-day variation were less than 10%. Conclusion The method might be simple, precise and sensitive and apply conveniently in studies of clinical pharmacy.
