帕金森病患者T淋巴细胞亚群及NK细胞的临床研究

目的研究帕金森病(PD)患者的T淋巴细胞亚群及NK细胞亚群变化,并探讨其临床意义。方法应用流式细胞仪直接免疫荧光染色法测定25例PD患者外周血T淋巴细胞亚群及NK细胞亚群,并与健康人组进行对照,同时对不同病期、不同伴随症状及L多巴治疗前后的患者进行相关分析。结果PD组CD3+、CD4+、CD8+、CD4+/CD8+水平较对照组降低,NK水平较对照组升高(P<0.05);病情重者CD3+、CD4+、CD4+/CD8+水平显著降低,NK水平升高(P<0.01);伴有抑郁和痴呆的患者CD3+、CD4+、CD4+/CD8+水平较非抑郁和痴呆患者下降(P<0.05);L多巴治疗前后T淋巴细胞亚群及NK细胞亚群水平无明显差异。结论PD存在T细胞免疫功能低下及NK细胞免疫平衡失调,病情重,伴有抑郁和痴呆的患者T细胞免疫功能更低。这对于进一步研究PD的病理生理机制及开辟新的防治途径有一定意义。

CLINICAL STUDY OF T LYMPHOCYTE AND NATURAL KILL CELL WITH PARKINSON DISEASE

Objective To study the changes and its clinical significance of T lymphocyte subsets and natural kill cells with Parkinson disease patients. Methords T lymphocyte subsets and natural kill cells were measured in 25 patients by the FCM direct immunefluorescent staining technique and compared with normal controls. Meanwhile patients were also compared with each other in different times, following symptoms, 12 of 25 cases were compared before and after administration of L-dopa. Results The levels of CD_3~+、 CD_4~+、 CD_8~+、 CD_4~+/CD_8~+ in PD were depressed, the levels of natural kill cells increased compared with healthy controls.(P<0.05) The levels of CD_3~+、 CD_4~+、 CD_4~+/CD_8~+ were decreased and NK were increased more significantly in severe patients.(P<0.01) The levels of CD_3~+、 CD_4~+、 CD_4~+/CD_8~+ were decreased in patients who were depressive and demential(P<0.05). The increase of T lymphocyte subsets and NK were insignificant after administration of L-dopa. Conclusions T lymphocyte immunefunctional decline and immune disturbance of NK were observed in PD patients. The levels of T lymphocyte subsets were even significantly lower in severe depressive and dementia patients, so this method is valuable to study the pathogenesis and pathology of PD for finding a new way of therapy and prevention.
