联合应用全反式维甲酸和化疗药物对肺腺癌细胞株A549细胞的作用

目的观察以不同方式联合应用分化诱导剂全反式维甲酸和化疗药物对肺腺癌细胞株A549细胞的作用,为临床诱导分化治疗肺癌提供实验基础。方法以1×10-6mol/L的全反式维甲酸作用于培养的肺腺癌细胞株A549细胞,1、2、3、5d后检测几种常用化疗药物对细胞的杀伤作用。同时观察同时加入1×10-6mol/L的全反式维甲酸和化疗药物对肺腺癌细胞株A549细胞的杀伤作用。结果同时应用全反式维甲酸和化疗药物时,对肺腺癌细胞株A549细胞的杀伤作用与单用化疗药物时差异无统计学意义。全反式维甲酸作用2d和3d时,化疗药物对肺腺癌细胞株A549细胞的杀伤作用强于全反式维甲酸作用前;全反式维甲酸作用5d时,化疗药物对肺腺癌细胞株A549细胞的作用较全反式维甲酸作用前强,但比3d时下降。结论先用全反式维甲酸作用后可增强肺腺癌细胞株A549细胞对化疗药物的敏感性,此作用与全反式维甲酸作用时间有关。不同联合用药方法的效果不同。

Combinative effect of all - trans retinolc acid with conventional anticancer agents on pulmonary carcinoma cell line A549

Objective To investigate the combinative effect of alltrans retinoic acid with conventional anticancer agents and provide experimental fundament for inducing differentiation of pulmonary carcinoma clinically.Methods All-trans retinoic acid and seven conventional anticancer agents were used in cultured pulmonary carcinoma cell line A549 in different ways.MTT was used to determine the combinative effect.Results After exposed to all-trans retinoic acid for 2 days,the sensitivityof A549 to NVB,MMC and DDP increased.And it increased slightly when exposed for 3 days,whereas decreased when exposed to all-trans retinoic acid for 5 days.When all-trans retinoic acid and anticancer agents were used together,the effect was not better than using anticancer agents alone.Conclusion All-trans retinoic acid may have synergistic effect with conventional anticancer agents.The effect is determined by the way of the agents were used.
