实验性变态反应性神经炎的大鼠淋巴细胞钾离子通道与细胞及体液免疫的关系

摘    要:目的:观察淋巴细胞K 通道在实验性变态反应性神经炎(EAN)大鼠中的表达情况及其与细胞和体液免疫的关联。方法:RT-PCR测定淋巴结和脾脏细胞Kv1.3和IKCa1的mRNA表达量,流式细胞分析相应细胞的增殖能力,间接ELISA测定特异性抗体水平,计算相关系数。结果:淋巴细胞Kv1.3和IKCa1的表达水平与细胞增殖能力紧密相关,与EAN临床严重程度无关,在一定程度上反应了EAN的总体细胞免疫水平。结论:淋巴细胞K 通道可以在以细胞免疫为主导的自身免疫性疾病中作为一个潜在的干预靶点。

Abstract:Aim: To observe the expression of lymphocyte voltage-gated K channel (Kv1.3) andintermediate-conductance calcium-activated K channel (IKCa1) in experimental allergic neuritis (EAN)rats and its correlations with cellular/humoral immunity. Methods: EAN was induced in Lewis rats withbovine peripheral myelin (BPM) and clinical severity was assessed. On day 9, 16, 23 and 30 post immunization,we evaluated the expression of mRNA for Kv1.3 and IKCa1 in lymphocytes from inguinal lymph nodes andspleens, the dividing cell proportions of two kinds of lymphocytes in different culture conditions, and thelevels of anti-BPM antibodies. Bivariate correlations were performed in statistical analysis. Results: ThemRNA expression of lymphocyte K channels was closely correlated with the capacity of lymphocyteproliferation but had little correlation with EAN clinical severity, which was able to reflect the cellularimmunity status of EAN on certain extent. Conclusions: Lymphocyte K channels may be a potentialtherapeutical target in autoimmune diseases mediated by cellular immunity.
