摘    要:显微切割人类染色体8q24.1带DNA的初级PCR产物与pUC19质粒载体进行体外连接,转化大肠杆菌DH5a,获得了329个白色克隆。已分析的53个克隆中,有36个单拷贝,这些单拷贝片段长度介于105bp~960bp间,平均350bp,它们与正常人基因组DNA杂交显示各自特异性,可作为8q24.1带的特异性遗传标记。

Abstract:The primary PCR products obtained from the human chromosome band 8q24.1 by mi-crodissection techniques were digested and ligated in vitro into plasmid pUC 19. After transforming Escherichia coli DH5 a with above ligation mixture, we had got 329 transfor-mants. The insert fragments from 53 randomly chosen transformants were labelled to hybridize with normal human genomic DNA digested with EcoRI or Hind I and 36 of them were determined as single-copy sequences. Electrophoresis analysis revealed that these single-copy sequences had an average insert size of 350bp with a range of 105-96bp. Furthermore, because each of them shows special hybridization-band patterns with normal human genomic DNA, they can be regarded as the genetic landmarks for the human chromosome band 8q24. 1.
