玻璃苣醇提取物对实验性哮喘豚鼠平喘作用机制的研究

摘    要:目的:观察玻璃苣醇提取物对实验性哮喘豚鼠平喘作用机制的实验研究。方法:将48只健康SPF级豚鼠随机分为空白组,模型组,地塞米松组,玻璃苣醇提取物高、中、低剂量组,灌胃给药,每日1次,连续7天,同时每天均于给药后30min,以组织胺激发诱喘。取肺灌洗液测定其一氧化氮(NO)含量,嗜酸性粒细胞(EOS)数目,干扰素-γ(IFN-γ),白细胞介素-4(IL-4)含量并计算其比值。结果:玻璃苣醇提取物使豚鼠肺灌洗液中EOS数目下降,NO浓度下降,IFN-γ含量升高,IL-4含量下降,比值升高。结论:玻璃苣醇提取物降低EOS在肺部的聚集,促进哮喘大鼠体内干扰素IFN-γ分泌和抑制IL-4表达,从而延长哮喘的潜伏期和降低支气管高反应性,达到治疗目的。

Abstract:To investigate the antiasthmatic mechanism of Borago alcohol extract on guinea pigs.Methods: Forty-eight healthy SPF-class guinea pigs were randomly divided into blank group,model group and dexamethasone group.Borago alcohol extracts were administered orally once a day for seven days at high,medium and low doses.Thirty minutes after drug administration each day,histamine was given to the animals to induce asthma.NO,eosinophils,IFN-γ,and IL-4 content of lung lavage fluid were measured,and the ratio was calculated.Results: In the lung lavage fluid of guinea pig,the amount of EOS decreased,the NO concentrations were decreased by Borago alcohol extract,while the amount of IFN-γ was increased.The amount of IL-4 was decreased,while the ratio of it was increased.Conclusion: The ethanol extract of Borago can reduce aggregation of EOS in the lungs,promoting asthma rats interferon IFN-γ secretion and inhibition of IL-4 expression,thereby it can prolong the incubation period of asthma and reduce bronchial hyperresponsiveness for therapeutic purposes.
