摘    要:目的研究彩虹明樱蛤多糖(MIP)对转染HBV的HepG2细胞表达功能的影响。方法以不同浓度(10-2~104μg/ml)MIP作用于体外培养的HepG2.2.15细胞,采用MTT法、时间分辨免疫分析法(TRFIA)和ELISA法检测MIP对HepG2.2.15细胞的毒性作用及HBsAg、HBeAg、Pre-S1Ag表达的影响,采用FQ PCR检测MIP对HBV DNA复制的抑制作用。结果 MIP在10-2~102μg/ml浓度范围内对HepG2.2.15细胞无毒性作用,TC50为4633μg/ml。MIP在10-2~102μg/ml浓度范围内,对HBsAg、HBeAg、Pre-S1Ag以及HBV DNA均有一定抑制作用,相应的治疗指数分别为129、28、20和83。结论 MIP具有一定的抗HBV活性。

Abstract:Objective To investigate the effects of Moerella iridescens polysaccharide(MIP) on Hepatitis B virus(HBV) transfected HepG2 cells.Methods HepG2.2.15 cells were treated with different concentrations of MIP(10-2–104μg/ml),and they were cultured in vitro.The MTT assay,time-resolved fluoreimmunoassay(TRFIA),and enzyme-linked immunosorbent assay(ELISA) were used to explore the cytotoxic effect of MIP on HepG2.2.15 cells and its effects on HBsAg,HBeAg,and Pre-S1Ag secreted by HepG2.2.15 cells.The fluorescence quantitative PCR(FQ-PCR) was employed to evaluate the inhibitory effects of MIP on the duplication of HBV-DNA.Results The results indicated that MIP had no clear cytotoxic effect on HepG2.2.15 cells in the concentrations from 10-2μg/ml to 102μg/ml,and the TC50 of MIP was 4,633μg/ml.MIP has some inhibitory effects in the concentrations from 10-2μg/ml to 102μg/ml on HBsAg,HBeAg,Pre-S1Ag,and HBV-DNA.The therapeutic indices(TI) of MIP on HBsAg,HBeAg,Pre-S1Ag,and HBV-DNA were 129,28,20,and 83,respectively.Conclusions MIP has a certain dose-dependent inhibitory effect on HBsAg,HBeAg,Pre-S1Ag,and HBV-DNA in the culture supernatant fluid of HepG2.2.15 cells;there is a dose–effect relationship,which shows that MIP has some anti-HBV activities.
