摘    要:目的探讨三氧化二砷(ATO)对MRL/lpr自发性狼疮小鼠脾脏单个核细胞(SMC)中DNA甲基转移酶1(DNMT1)和黏附分子淋巴细胞功能相关抗原-1(LFA-1)的亚基CD11a表达的影响及其可能作用机制。方法分离小鼠SMC,采用20g/ml植物血凝素和1000IU/ml白介素-2体外诱导培养SMC 48h后,随机分为空白组、ATO组和5-氮胞苷(5-Az)组,继续培养24h,采用RT-PCR法检测两组小鼠SMC中DNMT1及CD11a基因转录水平的表达情况。结果MRL/lpr小鼠ATO组中DNMT1-mRNA的灰度比值较5-Az组增高(P〈0.05),MRL/lpr小鼠ATO组中CD11a-mRNA的灰度比值较5-Az组降低(P〈0.05)。结论ATO治疗系统性红斑狼疮的作用机制可能与其能逆转低甲基化状态有关。

Abstract:Objective Toinvestigate the effect of arsenic trioxide(ATO) on the expression of DNA methyltransferase1 (DNMT1)mRNA and CD11a mRNA in splenic mononuclear cells (SMNC) of spontaneous lupus nephritis MRL/lpr mice.
