摘    要:目的 研究CDKN2 /p16基因缺失和蛋白丢失在肺癌发展中的作用。方法 采用免疫组化和改良标本处理的PCR技术 ,对 89例肺癌病人手术标本CDKN2 /p16基因第 1、2外显子纯合缺失和P16蛋白表达丢失情况进行分析对比研究。结果 CDKN2 /p16基因第 1或 (和 )第 2外显子总缺失率为2 5 8% (2 3/ 89) ,P16蛋白丢失率为 47 2 % (4 2 / 89) ,基因的缺失和蛋白的丢失集中发生于非小细胞肺癌(NSCLC) ,并与转移和分期有关。结论 CDKN2 /p16基因的异常可能是NSCLC区别于小细胞肺癌(SCLC)的特有的分子事件 ,其在NSCLC的发生发展中起一定的负调控作用

Abstract:Objective To study CDKN2/p16 gene homozygous deletion and p16 protein loss in the genesis and progression of lung cancer. Methods Improved multiplex PCR technique and immunohistochemistry were applied to detect deletion of CDKN2/p16 gene exon1 and exon2 and loss of p16 protein in the 89 cases of lung cancer. Results Total rate of exon1 or/and exon2 deletion was 25 8%(23/89), and p16 protein loss rate was 47 2%(42/89) in lung cancer. CDKN2/p16 gene deletion and protein losses occurred in NSCLC, and were related to metastasis and progressive stage. Conclusion Abnormality of CDKN2/p16 gene is a specific molecular event in NSCLC, and may play an important role in the progression of NSCLC.
