摘    要:目的探讨姜黄素对TNF—α诱导人脐静脉内皮细胞产生NO以及表达ICAM-1的影响。方法体外培养人脐静脉内皮细胞,用10ng/mLTNF—α刺激后,再加入(5~40)μg/mL姜黄素共同孵育lh,采用MTT和ELISA法检测姜黄素对内皮细胞的增殖以及N0和ICAM-1的表达。结果(5~40)μg/mL姜黄素处理HUVEC细胞后,能明显抑制ICAM-1的表达,并增加NO的产生,呈剂量依赖性。结论姜黄素通过抑制内皮细胞ICAM-1的表达,促进NO的分泌,从而发挥内皮细胞保护作用以及抗动脉粥样硬化。

Abstract:[Abstract] Objective To study the effect of cnrcumin on HUVECs production of NO and expression of ICAM-1. Methods The HUVECs were cultured in vitro and stimulated by 10ng/mL TNF-α , then incubated with (5 - 40 ) μg/mL of curcumin for lh. MTT and ELISA was used to detect NO and ICAM-1 production. Results ICAM-1 expression was inhibited by (5 - 40)μg/mL of curcumine, and increased NO production in dose-dependent manner. Conclusion Curcumin exhibit cytoprotection and antiartherosclerosis effect may mediated by inhibition of ICAM-1 expression and NO production.
