摘 要: | 目的探讨加味温胆汤对抑郁模型大鼠海马NMDA-NR1mRNA/蛋白表达的影响作用。方法以孤养结合慢性不可预见性应激抑郁模型大鼠为研究对象,在实验7d、14d、21d、28d时,检测各组大鼠旷场实验中水平运动、垂直运动得分,并采用QRT-PCR、Western-Blot技术检测各组大鼠海马NMDA-NR1mRNA/蛋白表达变化。结果 21d、28d时,与空白组比较,模型组水平运动、垂直运动得分降低(P<0.01);与模型组比较,中、西药组水平运动、垂直运动得分增加(P<0.05或P<0.01)。21d、28d时,与空白组比较,模型组大鼠海马NMDA-NR1mRNA/蛋白表达增多(P<0.01);与模型组比较,中、西药组大鼠海马NMDA-NR1mRNA/蛋白表达减少(P<0.05或P<0.01)。结论加味温胆汤的抗抑郁效应可通过阻抑海马NMDA-NR1mRNA/蛋白表达而实现。

Abstract: | Objective To explore the effect of Modified Wendan decoction on N-methyl-d-aspartic acid(NMDA)-NR1mRNA/protein expression in the hippocampus of the rat model with depression.Methods The rat models with unpredictable chronic stress and isolation-induced depression were studied.Models were randomly divided into four groups and the rats of each group were intragastrically perfused with corresponding interventions at the day 7th,14th,21st,and 28th.The behaviors of open-field test were examined and the NMDA-NR1mRNA/protein expression in the hippocampus was tested by the method of QRT-PCR and Western-Blot.Results At the day 21st,28th,the score of vertical motion and horizontal motion behaviors of open-field test in model group was lower than that in blank group(P<0.01).The score of vertical motion and horizontal motion behaviors of open-field test in Chinese medicine group and western medicine group was higher than that in model group(P<0.05or P<0.01).At the day 21st and 28th,The expression of NMDA-NR1mRNA/protein in the hippocampus of the rats in model group were higher than that in blank group(P<0.01).The expression of NMDA-NR1mRNA/protein in the hippocampus of the rats in Chinese medicine group and western medicine group were lower than that in model group(P<0.05or P<0.01).Conclusion The mechanism of the antidepressant effect of Modified Wendan Decoction is related to down-regulate the expression of NMDA-NR1mRNA/protein in the hippocampus of depression. |
