摘    要: 目的 研究股骨头坏死(osteonecrosis of the femeral head,ONFH)患者滑膜组织来源的类成纤维细胞对人Burkitt淋巴瘤(Burkitt lymphoma,BL) Raji细胞株增殖的影响。方法首先分离和培养来自ONFH患者滑膜组织的原代类成纤维细胞,随后将其与Raji细胞共培养;用细胞计数试剂盒(CCK 8)检测Raji细胞的增殖情况;用实时定量PCR(quantitative real time PCR,qRT PCR)检测Raji细胞中P53、P21及CD9基因的表达水平。结果ONFH滑膜来源的细胞为贴壁的类成纤维细胞。共培养组中的Raji细胞增殖速率显著高于单独培养组(只培养Raji细胞)。相对于单独培养组,共培养24 h后,P53、P21及CD9的表达量分别为0.82(P<0.05)、1.75(P<0.05)及1.99(P<0.05),48 h后,则分别为0.72(P<0.05)、1.03及0.56(P<0.05)。结论在共培养条件下,ONFH患者滑膜组织来源的类成纤维细胞可能通过调控Raji细胞中P53、P21及CD9的表达来促进Raji细胞的增殖。

Abstract:Objective  To investigate the effect of fibroblast-like cells which derived from the synovial membrane of the patients with osteonecrosis of the femeral head (ONFH) on the proliferation of Raji cells from human Burkitt lymphoma (BL).Methods  Primary fibroblast-like cells derived from the synovial membrane of ONFH patients were isolated and cultured,and then a co-culture system was established with Raji cells.The proliferation of Raji cells was determined by CCK-8 assay,and the expressions of P53,P21 and CD9 mRNA in Raji cells were determined by quantitative real-time PCR (qRT-PCR).Results  The cells derived from the synovial membrane of ONFH were adherent fibroblast like cells.The proliferation rate of Raji cells in co-culture group was significantly higher than that in individual group (cultured Raji cells alone).Compared with the individual group,the relative expression amount of P53,P21,and CD9 mRNA after co-cultured for 24 hours in co-culture group was 0.82 (P<0.05),1.75 (P<0.05),and 1.99 (P<0.05),respectively; and was 0.72 (P<0.05),1.03,and 0.56 (P<0.05),respectively after co-cultured for 48 hours.Conclusions  Under co-culture condition,the fibroblast-like cells from synovial membrane of ONFH patients could significantly promote the proliferation of Raji cells by regulating the expression of P53,P21,and CD9 mRNA.
