摘    要:目的 观察三氧化二砷(ATO)对支气管哮喘小鼠Th 17细胞和IL-17的影响.方法 30只BALB/c小鼠随机分为对照组、哮喘组和ATO组.末次激发24小时后,测定气道肺阻力,收集支气管肺泡灌洗液(BALF),细胞分类计数,流式细胞仪检测脾脏Th17细胞阳性率.ATO干预20小时后,检测培养上清液及BALF中IL-17及IL-4水平.结果 ①哮喘组BALF中各类炎症细胞均明显高于对照组(P<0.05),而ATO组明显低于哮喘组(P<0.01).②哮喘组肺阻力、脾脏CD4+T细胞总数、Th17细胞阳性率、BALF中IL-17及IL-4均明显高于对照组,而ATO组上述指标均明显低于哮喘组(P<0.05).③ATO干预组培养上清液中IL-17、IL-4含量显著低于原液组(P<0.01).④Th17细胞阳性率、IL-17含量均与中性粒细胞数量呈显著正相关(P<0.05).结论 ATO能够抑制Th17细胞、IL-17的表达,减轻哮喘小鼠气道炎症及高反应性.

Effects of arsenic trioxide on Th17 cells in asthmatic mice

Abstract:[Abstract] Objective To investigate the effect of arsenic trioxide(AT()) on the Thl7 cells and IL-17 in asthmatic mice. Methods 30 BALB/c mice were randomly divided into control group, asthma group and ATO group. 24 h after the last challenge, lung resistance was measured, BALF was collected to classify and count, the positive ratio of Thl7 cells was detected by FCM. 20 h after ATO treatment, the levels of IL-17 and IL-4 in BALF and culture supernatants were quantified. Results The numbers of all inflammatory cells in BALF were significantly higher in asthma group com- pared with control group, and were significantly lower in ATO group compared with asthma group(_P,Q0.01). Lung re- sistance, the total number of CD4 + T cells, the1 positive ratio of Th17 cells and the level of IL-17 and IL-4 in BALF were significantly increased in asthma group compared with control group, and were significantly decreased in ATO group compared with asthma group(P~0. 05). The level of IL-17 and IL-4 in culture supernatants was lower in AT() induced group compared with control group(P%0.01). (4) The positive ratio of Th17 cells and IL-17 level positively correlated with the number of neutrophils (P%0.05). Conclusion Arsenic trioxide can inhibit the expression of the Thl7 cells and IL-17, and reduce airway inflammation and reactivity in asthmatic mice.
