氧化苦参碱对大鼠缺血再灌注心肌损伤生化指标的影响

摘    要:目的:探讨氧化苦参碱对大鼠缺血再灌注心肌的保护作用.方法:45只成年大鼠随机分成3组:正常对照组、缺血再灌注组、氧化苦参碱组,麻醉大鼠后冠脉结扎30 min,再灌60 min后,各组立即取出心脏,用冰盐水冲洗、吸干、称重后以生理盐水为介质,用匀浆器制成心肌组织匀浆,观察3组大鼠心肌组织匀浆中脂质过氧化物质、酶学指标的变化.结果:氧化苦参碱组超氧化物歧化酶、谷胱甘肽过氧化物酶活力、一氧化氮水平、诱导型一氧化氮合酶活力显著高于缺血再灌注组,丙二醛显著低于缺血再灌注组(P<0.01).结论:氧化苦参碱对大鼠缺血再灌损伤的心肌具有保护作用,此作用可能与抑制心肌脂质过氧化、增强抗氧化能力有关.

Abstract:Aim: To observe the effect of oxymatrine (OMT) on myocardial ischemic reperfusion (IR) injury in the rat heart in vitro.Methods:Forty-five adult Wistar rats were randomly allocated into three groups: normal control groups, IR group, and OMT group. The myocardial ischemia was induced by coronary artery ligation for 30 min and reperfusion for 60 min. Myocardial NO, malondialdehyde(MDA), the activities of NOS,superoxide dismutase(SOD), and glutathione peroxidase(GSH-PX) were determined. Results: Compared with IR group, the levels of myocardial SOD, GSH-PX, NO, and NOS were significantly higher while the level of MDA was lower in OMT group (P<0.01). Conclusion: OMT plays a protective role in rat myocardial IR injury in vitro which may involve in its inhibiting lipid peroxidation and enhancing anti-oxidation.
