摘    要:本实验用双波长紫外分光光度法测定20名健康人服用氨茶碱后的血浆浓度并计算其药动学参数.结果显示,服用氨条碱0.3g后4h时血浆浓度超过10mg/L者4人,其余均在5~10mg/L之间,而8h时有3人的氨茶碱血浆浓度低于3.5mg/L。20名受试者的血浆半衰期波动范围在1.88~10.94h之间,其中5人在6~9h之间,而12人的半衰期低于6h.说明服用同等量的氨茶碱后个体血浆浓度及半衰期差异甚大。因此在应用氨茶碱时应常规测定血浆浓度,以调整用量达到个体化治疗。

Study on pharmacokinetics of aminophylline in healthy volunteer

Abstract:The plasma levels of aminophylline of 20 healthy volunteer subjects were determined with uvspectrophotoometry and the pharmacokinetic parameters were calculated in these subjects. The results showed that at 4h after 0. 3g aminophylline token orally, there were 4 healthy subjects with more than 10mg/L of aminophylline in plasma, the others were between 5 and 10mg/L, and there were also 3 healthy subjects with less than 3. 5mg/L at 8h. The range of half-life time(T)varied from 1. 88 to 10. 94h in the 20 healthy subjects. Among them 5 subjects were between 6 and 9h and the T in 12 subjects was less than 6h. The results explain that the effect and adverse reaction of treating with aminophylline relate to individual reaction. In same dosage, some people may appear in toxic reaction, others in no effect. In this condition, in addition to diffcrentiation of T of aminophylline, plasma levcl of aminophylline will markedly differ after taken consecutively. Thus, plasma levels of aminophylline should be routinely determined to adjust the dosage of aminophylline for individualizing treatment.
