摘    要:目的:建立忍冬藤双波长融合测定方法,同时测定忍冬藤中绿原酸、咖啡酸、马钱苷含量,该方法稳定可靠,可以用来全面控制忍冬藤药材质量。方法:采用高效液相色谱法,Agilent TC-C18(4.6mm×250mm,5m)色谱柱;以0.3%甲酸水、乙腈为流动相体系进行梯度洗脱;流速1mL/min;柱温25℃;进样10μL;检测波长:236nm、327nm;使用Matlab7.1编程进行谱图融合。结果:方法学考察结果良好,绿原酸在0.5212~2.4757μg/mL范围内线性关系良好;咖啡酸在0.1960~0.9310μg/mL范围内线性关系良好;马钱苷在0.5760~2.7360μg/mL浓度范围内线性关系良好,平均回收率分别为99.60%(RSD=0.76%),99.73%(RSD=0.60%),99.48%(RSD=0.73%)。结论:双波长融合法为忍冬藤药材提供了合理、可靠的质控方法,可以全面、整体地评价忍冬藤药材及含有忍冬藤药材的制剂质量。

Abstract:Objective:Dual-wavelength fusing method was establisheet for simultaneous determination of chlorogenic acid,Caffeic acid and Loganin,which is stable and reliable and can be used as a comprehensive quality control for Caulis Lonicerae.Methods:By HPLC,The separation was performed on an Agilent TC-C18 column(4.6mm×250 mm,5 m);acetonitril-0.3% methanoic acid was used as the mobile phase,with the flow rate of 1.0mL·min-1;the temperature was maintained at 25℃;the volume was 10μL,the detection wavelength was set at 236nm and 327nm,using Matlab7.1 programmation to fuse the chromatograph chart.Results:Methodology of the investigation results is qualified,chlorogenic acid show good linear relationship in the range of 0.5212~2.4757μg/mL;Caffeic acid show good linear relationship in the range of 0.1960~0.9310μg/mL;Loganin show good linear relationship in the range of 0.5760~2.7360μg/mL;the average recovery was 99.60%(RSD=0.76%),99.73%(RSD%=0.60%),99.48%(RSD=0.73%).Conclusion:Dual-wavelength fusing method supply a Reasonable and dependable quality control method to evaluate the quality of Caulis Lonicerae or the preparation with it all sidely and integrally.
