丹参注射液对急性肺损伤模型大鼠血管生成素表达的影响

摘    要:目的:研究丹参注射液对急性肺损伤模型大鼠血管生成素(Ang-1)-1、Ang-2表达的影响。方法:大鼠尾静脉注射脂多糖(5mg·kg^-1)复制急性肺损伤模型。实验分为3组,即正常(生理盐水1mL)、模型(生理盐水1mL)和丹参(10g·kg^-1)组。给药12h后处死大鼠,收集肺泡灌洗液、血液、肺组织进行肺湿重/干重(W/D)值、肺系数(LI)、肺通透系数(LPI)等的计算;HE染色观察大鼠肺组织病理改变;Westernblot法测定Ang-1、Ang-2在肺组织的表达。结果:与正常组比较,模型组大鼠肺组织W/D比值、LI、LPI显著升高(P〈0.05),且病理切片都有明显的炎症损伤改变,Ang-1表达明显减弱,Ang-2表达明显增强。与模型组比较,丹参组大鼠肺组织W/D、LI、LPI显著降低(P〈0.05),Ang-1表达明显增强,Ang-2表达明显减弱。结论:丹参注射液对急性肺损伤有一定保护作用,其机制可能与Ang-2、Ang-1的表达调控有关。

Abstract:OBJECTIVE: To study the effects of Danshen injection on the expression of Ang-1 and Ang-2 in rats with acute lung injury. METHODS: Acute lung injury model rats were induced by lipopolysaccharide (5 mg·kg-1)via tail vein. Model rats were divided into 3 groups, i.e. normal group(normal saline 1 mL), model group(normal saline 1 mL) and Danshen injection group(10 g·kg-1). Those model rats were sacrificed 12 h after medication. Bronchoalveolar lavage fluid, blood samples and lung tissue were collected, and lung wet/dry (W/D) weight ratio, lung index (LI) and lung permeability index (LPI) were calculated. The pathological changes of lung tissue in rats was observed by HE staining; the expressions of Ang-1 and Ang-2 in lung tissue were determined by Western blot assay. RESULTS: Compared with normal group, W/D weight ratio, lung index (LI) and lung permeability index (LPI) were increased significantly(P0.05), and pathological section showed that there was obvious inflammatory injury in model group. The expression of Ang-1 decreased significantly, while that of Ang-2 increased significantly. Compared with model group, the W/D, LI and LPI of Danshen injection groups decreased significantly, and the expression of Ang-1 increased significantly while that of Ang-2 decreased significantly. CONCLUSION:Danshen injection have protective effect against acute lung injury, which is associated with the expression of Ang-2 and Ang-1.
