归元片对吗啡条件性位置偏爱的影响

目的:探讨不同剂量的戒毒中药归元片对吗啡诱导的条件性位置偏爱(conditionedplacepreference ,CPP)的影响及其自身是否引起CPP。方法:(1)♂Wistar大鼠sc吗啡(5mg·kg-1)训练7d ,干预组于每次给吗啡前15min注射不同剂量的归元片(12 . 5 - 37 .5mg·kg-1,sc) ,观察其对吗啡CPP效应的干预作用;(2 )♂Wistar大鼠sc归元片(2 5mg·kg-1及5 0mg·kg-1)训练7d ,d8测定大鼠对伴药侧的偏爱效应。结果:(1) 5mg·kg-1吗啡可以诱导大鼠对伴药侧产生显著的CPP ;而两个剂量的归元片均不能使大鼠形成CPP ;(2 ) 2. 5mg·kg-1和37 .5mg·kg-1的归元片可剂量依赖性地降低吗啡诱导的CPP的形成。结论:一定剂量的归元片可拮抗大鼠对吗啡偏爱效应的获得,且自身不引起CPP ,提示该药有治疗阿片类药物精神依赖性的潜力。

THE EFFECT OF GUIYUAN TABLETS ON MORPHINE- INDUCED CONDTIONED PLACE PREFERENCE

Objective: To study the effect of different doses of Guiyuan tablets on morphine induced conditioned place preference(CPP) and observe whether Guiyuan tablets itself would induce CPP. Methods:(1) Male Wistar rats were administered morphine (5 mg·kg -1, sc) for 7 days to observe Mor CPP; different doses of Guiyuan tablets(12.5-37.5 mg·kg -1 were given (sc) 15 min before the administration of morphine to see the effect of Guiyuan on Mor induced CPP; (2) Two groups of male Wistar rats were trained separately with Guiyuan tablets(25 mg·kg -1 and 50 mg·kg -1, sc)for 7 days, and on d8,they were tested whether Guiyuan tablets could induce CPP. Results: (1)5 mg·kg -1 morphine (sc) induced rats obtaining a significant place preference. And both doses of Guiyuan tablets did not induce CPP; (2) 25 mg·kg -1 and 37.5 mg·kg -1 administered (sc) before morphine administration blocked the development of morphine-induced CPP in a dose dependent manner. Conclusion:Guiyuan tablets can meddle in reinforced effect induced by morphine at the doses that cannot induce CPP,which suggests that Guiyuan tablets might be useful in the treatment of opiate psychological dependence.
