人膀胱癌组织中SOD同工酶的表达及其生物学意义

摘    要:目的:探索人膀胱癌组织中SCD同工酶的表达状态及其生物学意义。方法:选取经过病理学证实的人膀胱癌组织及邻近癌旁组织,并以非膀胱癌患者的膀胱黏膜组织作对为对照,分别用硫代巴比妥酸法和亚硝酸盐法测定组织中的丙二醛(MDA)含量、超氧化物歧化酶同工酶(CuZnSCD、MnSCD)活力。结果:按照正常膀胱黏组织—癌旁组织—癌组织的发生发展过程,组织中MDA含量依次显著升高,而SOD同工酶活力(尤其MnSCD活力)依次显著下降。且随肿瘤临床分期和病理分级的递增而分别升高和降低。结论:膀胱癌的发生发展与组织中氧化应激的亢奋及抗氧化酶活力的损伤密切相关,且这一变化早于组织学变化。

Abstract:Objective:To explore the status of expression of SOD isoenzyme in human bladder carcinoma tissues and its biological significance. Methods:The contents of malonaldeyde (MDA) and the activities of superoxide dismutase isoenzyme including CuZnSOD and MnSOD in bladder cancerous tissues, cancer-free adjacent cancerous tissue and normal bladder mucosa tissues (as control) were assayed with thiobarbituric and nitrite method, respectively.Results:According to the group sequence: the control tissue group, cancer-free adjacent cancerous tissue group and cancerous tissue group, the levels of MDA increased gradually and the activities of SOD isoenzyme (especially MnSOD) decreased significantly. In addition, the MDA levels and SOD isoenzyme activities in bladder cancer tissues increased and decreased respectively with the increasing of tumor grades and tumor stages. Conclusions:The carcinogenesis of human bladder is closely related to the enhancing of oxidative stress and the decline of antioxidase activities, which occurs earlier than the changes of histology.
